you may be able to bring the VIN to your local Lexus dealer and have them look up the service history. I think routine maintenance is going to tell you a lot more about the car than trying to crawl under it and look at the drive train. If you're going to "kick the tires" so to speak. I'd check under the spare tire cover for water damage, be sure the fluids are level and don't smell burnt and look at the paint. If you see small particles under the the paint, that means body work.. BEWARE....

Have the front axles, CV joints, suspension parts completely inspected. The GS had some issues with the suspension parts breaking down prematurely.

make sure each panel {door, fender, trunk all have vin tags. If not it was replaced. Have them get you a copy of the service history from the local lexus dealer. its nationwide so they can tell if its been done anywhere. check the outer edges of the tires for alignment issues. the car has a 6yr 70k powertrain warranty. so the big stuff you may be able to checked.

Same as any other used car. Drive it and be sure you like it and everything works. Do the simple stuff yourself like ensuring the interior is clean and usually free of wear its six years old there will be some on the seats and carpet. be sure the tires are good wear, good tread left . Have a mechanic not the dealer that you trust check the engine, transmission, axles and suspension out for any sign of a problem or if everything looks brand new may have been abused or wrecked . Carfax is useful too.

it's a luxury car and lexus is really good so all you have to ask them is when you may want to bring it in. as well, you could leave it at the dealer for about an hour or so and ask them to look at all the basic things and and ask them to fix stuff that is not working.
