I have a 1964 and 1/2 Mustang. 289 engine, and a four speed manual transmission. My problem is, that when I go to manually turn the engine over, counter clockwise, the back of the block, or bell housing makes a deafening "clicking" sound. This a serious concern of mine, considering that the transmission and pressure plate are lined up exactly as they should. WHAT DO I DO.?..

Have you tried unbolting the starter, then turning the motor over? You may have a stuck drive in the starter, that contacts the teeth of the flywheel when turning.

I could drop the tranny and see exactly where the sound is coming from. Hopefully something broke doesn't fall out. despite, you'll sooner or later have to go in and learn what is causing it

sounds to me one of your flywheel bolts are loose check all the fltwheel bolts before you must replace the flywheelor you have a bad transmission converter

Try jacking up the back wheels, start it, put it into gear and see if it makes the sound.

If then take it separately and look for loose/missing bolts, pieces of gear like I found in my '65 mustangs rear end because some jerk off did not put a bearing into the casing and it snapped a part of the case off and destroyed the ring gear

If it does not make it when it's under power, then what you're hearing may be normal.

Can't say the problem, but the engine turns clockwise, why are you turning it the reverse way the engine turns?
