Most hybrids use CVT Continuously variable transmissions which are far more fuel economical than regular transmissions with overdrive.

Always use overdrive to get better gas mileage in hybrid or non hybrid.

depends on you are handling
if you have long runs on the highway then the difference will be fair
in the city it'll be less

Well, considering a that a hybrid performs with better MPG after reaching "speed", then no, it could not make lots of difference. I mean most hybrids run gas till they reaching cruising speed and switch to electric. , if the transmission offered an overdrive or not could be a mute point in mpg consideration. As for city driving, with the stop and go, there is not a whole lot of electric going on yes, overdrive could seem right in this enviroment. As for towing/cargo carrying, well, I do not use the overdrive on my Jeep, the manual claims that it'll because to much stress on the engine and transmission. could be the same on a Hybrid.
